高效液相色谱-蒸发光散射检测法测定归芪口服液中黄芪甲苷含量

HPLC-ELSD determination of astragaloside in chinese medicine Guiqi oral solution

目的建立高效液相色谱-蒸发光散射检测法测定归芪口服液中黄芪甲苷的含量。方法采用色谱柱：Thermo BOS Hypersil C18（5μm,250mm×4.6mm）;流动相：乙腈-水（35∶65）;流速：1.0mL·min^-1;ELSD参数：漂移管温度90℃,N2气体流量：1.6L·min^-1。结果黄芪甲苷在0.6～15μg范围内线性关系良好（r=0.9994）,3个浓度的平均回收率分别为96.9%,97.0%,98.2%;RSD分别为2.0%,1.9%,1.7%。结论本方法操作简便、准确,重复性好,可有效控制归芪口服液中黄芪甲苷的内在质量。

OBJECTIVE To develop an HPLC-ELSD method for determination of astragaloside Ⅳ in chinese medicine Guiqi oral solution.METHODS Using Thermo BDS Hypersil C18（5μm,250×4.6mm） with acetonitrile-water（35∶65） as the mobile phase,1.0mL·min^-1 as the flow rate,90℃ as inlet heater,1.6L·min^-1 as N2 atmosphere flow.RESULTS The linear range was 0.6～15μg with correlation coefficient of 0.9994 and different recovery of 96.9%、97.0%、98.2%,the RSD of them were 2.0%,1.9%,1.7%.CONCLUSIONS This method is simple,reproducible,good sensitive and it can be used for the quality control of astragaloside Ⅳ in Guiqi oral solution.
