摘要
目的探讨活血化瘀复方中药芪丹通脉片(QDTMT)对缺氧模型大鼠血清血管内皮生长因子(VEGF)浓度的影响及其内皮保护作用的相关分子机制。方法将大鼠随机分为正常对照组、低氧+QDTMT组(QDTMT组)、低氧+生理盐水组(生理盐水对照组),采用低压低氧法建立大鼠缺氧模型,并对肝脏组织进行病理形态学分析;分别于缺氧1周后、缺氧2周后用ELISA试剂盒检测各组大鼠血清中VEGF含量。结果造模后,QDTMT组病理形态学损伤较轻。缺氧1周后,QDTMT组大鼠血清中VEGF含量明显高于正常对照组和生理盐水对照组;缺氧2周后,QDTMT组大鼠VEGF含量仍高于正常对照组,但明显低于生理盐水对照组。结论缺氧后QDTMT对VEGF分泌的双向调节作用,可能为其内皮保护机制之一。
Objective: To find the protective molecular mechanisms of TraditionalChinese Medicine, and to study the effects of Qidantongmai Tablet(QDTMT) on the expression level of VEGF and KDR in rats with hypoxie model. Methods Rats were randomly divided into 3 groups normal, treatment and control. Before analysis the liver pathological changes, the rats were exposed to low pressure and low oxygen conditions in hypobaric and hypoxic chamber for the establishment of animal model. After 1 week and 2 weeks of bypoxia, concentrations of VEGF in rat serum were detected. Results The liver pathological changes decreased in treatment group. After 1 week of hypoxia, that the concentration of VEGF in serum of treatment group was higher than that of control group and normal group, After 2 weeks of hypoxia, the VEGF level of control group was higher than that of treatment group. Conclusion Two, ways regulation effect on VEGF level may be one of the mechanism for QDTMT to protect the endothelial tissue in hypoxia condition.
