摘要
通过氨气敏电极对脲酶催化反应中所生成氨的跟踪,研究了催化反应的动力学响应和稳态响应的规律。结果表明,电位法测定所需要的脉酶浓度可低达0.65mg/ml 左右;最适宜温度为25℃;电极响应速度最快的 pH 值为8.9;在不同 pH 下,稳态电位(E)-log[尿素]的线性范围和灵敏度也不相同,在 pH 7.3时,线性范围为3.0×10^(-4)~2.0×10^(-2)mol·l^(-1)。
In this paper,the effects of urease concentra-tion,urea concentration,temperature and pHvalue on the characteristics of both the kineticresponse and steady-state response of electrode wasexplored by following the tracks of ammoniaproduced from the enzymatic hydrolysis reactionof urea.The urease concentration needed is as lowas 0.65 mg/ml.The optimum temperature of thereaction is about 25℃.The pH value at which theelectrode shows supreme steady-state response is8.9.The linear range and the sensitivity betweensteady-state potential(E)and log[urea]aredifferent at various pH values.At pH 7.3,thelinear range is 3.0×10^(-4)~2.0×10^(-2)mol·l^(-1).
