摘要
在Sprague-Dawley大鼠过敏性气道炎症模型上观察不同药物的抗炎作用。ip地塞米松(0.5mg·kg-1),粉防己碱(30mg·kg-1)以及非肽类神经速激肽受体拮抗剂CP-96345和SR-48968(各1mg·kg-1),每日2次,连续3d,可抑制卵白蛋白1mg致敏大鼠吸入抗原后6h或24h肺灌洗液中的白细胞数量增多,明显减轻细支气管和小血管周围嗜酸性细胞浸润及管壁水肿等炎症状况。结果证明大鼠模型可以作为评价平喘药作用的工具,并提示速激肽参与气道过敏性炎症。
The anti inflammatory effects of various drugs were observed in a Sprague Dawley (SD) rat model of airway allergic inflammation. Intraperitoneal injections of dexamethasone (0.5 mg·kg 1 ), tetrandrine (30 mg ·kg 1 ) and a combination of nonpeptide tachykinin receptor antagonists, CP 96345 and SR 48968 (1 mg·kg 1 , respectively), twice a day for 3 d, significantly decreased the ovalbumin (OA, 1 mg) aerosol induced increases in leukocyte numbers in the bronchoalveolar lavage fluid, edema of airway walls, and infiltration of eosinophils around the bronchioles and vessels 6 and 24 h after inhalation in the OA sensitized SD rats. This result indicates the usefulness of SD rat allergic airway inflammation as a model for evaluation of the anti inflammatory effects of anti asthmatic drugs, and the involvement of endogenous tachykinins in the allergic airway inflammation.
