Unsymmetrical dimethyl hydrazine (UDMH) is a rocket propellant. It has been identified as a potential human carcinogen by NIOSH,but most mutagenicity tests show negative results. We analyzed the ability of UDMH to induce aneuploidy in CHL cells with chromosome enumeration and cytokinesis blocked (CB) micronucleus assays. In slide preparation, the cell pellet was treated with 94 mmol·L 1 KCl hypotonic solution for 2-3 min to preserve cytoplasm. UDMH caused significant increases in the frequencies of aneuploidy at concentrations of 15-60 mg·L 1 , but the incidence of chromosomal aberrations showed no difference between the treated group and normal control. UDMH also markedly increased the frequencies of micronuclei in binucleated cells. These results suggest that UDMH may have ability to induce aneuploidy in cultured cells.
