生长因子对人精原干细胞体外存活和增殖的影响 被引量:3

摘要 目的:探讨3种生长因子——干细胞因子(SCF)、白血病抑制因子(LIF)和碱性成纤维细胞生长因子(bF-GF)对精原干细胞(SSCs)体外存活及增殖的作用。方法:在培养液中加入不同浓度的SCF、LIF和bFGF,各生长因子均设浓度为0、5、10、20μg/L4个浓度组,每种生长因子不同浓度1式3份,每隔8~12h观察细胞的体外存活及增殖情况。应用光镜、电镜观察SSCs的形态结构特点。结果:培养液中加入不同浓度的SCF、LIF及bFGF后,SSCs的存活时间均比对照组有所延长,组间比较差异有显著性(P<0.05)。结论:生长因子SCF、LIF和bF-GF等可促进SSCs体外存活及增殖。 Objective: To study the effects of growth factors on the survival and proliferation of human spermatogonial stem cells (SSCs) in vitro. Methods : SSCs were treated with the growth factors SCF, LIF and bFGF added to the culture, each at the concen- trations of 0, 5, 10 and 20 ttg/L and repeated three times. The survival time and proliferation rate of the cells were determined every 8-12 hours and their morphological features observed with the light microscope and electron microscope. Results: The survival time and proliferation rate of the SSCs were significantly increased in the treated groups as compared with the control ( P 〈 0. 05 ).
