摘要
目的研究炎性因子白细胞介素-6(IL-6)和白细胞介素-1β(IL-1β)对人冠状动脉平滑肌细胞(HCASMC)分泌妊娠相关血浆蛋白A(PAPP—A)的影响。方法一期:分别加入20μg/L的IL-1β(IL-1β组)、10μg/L的IL-6(IL-6组)和0μg/L的炎性因子(对照组),孵育2、4、8、24、36h后,分别收集细胞上清液;二期:分别采用IL-1β(0、5、20、40μg/L)和IL-6(0、5、10、50μg/L)刺激HCASMC,6h后收集细胞和细胞上清液。采用ELISA法检测细胞上清液内PAPP—A的表达量。结果一期:IL-1β组PAPP—A表达量在2h时开始增加,8、24、36h其浓度显著高于对照组,并随时间的延长而不断增加;IL-6组PAPP—A的表达量在2h开始增加,4、8、24、36h其浓度显著高于对照组,并随时间的延长而不断增加。二期:随着IL-1β和IL-6剂量的增加,PAPP—A的表达量不断升高,其中TL-1β组20μg/L和40μg/L均显著高于0μg/L时的浓度;IL-6组10μg/L和50μg/L均显著高于0μg/L时的浓度。结论IL-1β和IL-6可使HCASMC分泌PAPP—A增加,并呈时间和剂量依赖性。
Objective To observe the effect of IL-1β and IL-6 on secretion of pregnancy-associated plasma protein A(PAPP-A) in humam coronary atery smooth muscle cell(HCASMC). Methods At the first stage, HCASMCs were incubated with IL-1β (20 μg/L), IL-6 (10 μg/L) and inflammation factor(0 μg/L,served as the control group). Culture media were collected after 2, 4, 8, 24 and 36 h. At the second stage: HCASMCs were stimulated by IL-1β(0, 5, 20, 40 μg/L) and IL-6(0,5,10,50 μg/L);0 μg/L served as the control group. Culture media were collected after 6 h. The concentrations of PAPP-A in culture media were measured by ELISA. Results At the first stage,incubation of HCASMC with 20 μg/L of IL-1β and 10 μg/L of IL-6 resulted in increase in secretion of PAPP-A which increased continuously with time. In the group cultured with IL-1β,it began to increase at 2 h, and the concentrations at 8,24 and 36 h were higher than those in the control group significantly. In the group cultured with IL-6, it began to increase at 2 h,and the concentrations at 4, 8,24,36 h were higher than those in the control group significantly. At the second stage,incubation of HCASMC with different concentrations of IL-1β and IL-6 caused dosedependent increase in PAPP-A secretion. In the group cultured with IL-1β,the PAPP secretion in both 90 μg/L and 40 μg/L groups exceeded the control group significantly. In the group cultured with IL-6,the PAPPsecretion in both 10 μg/L and 50 μg/L groups exceeded the control group significantly. Conclusions IL-1β and IL-6 can induce the secretion of PAPP-A by HCASMCs,implicating that inflammation may play an important role in the development of ACS.
