镰刀菌诱导后苦瓜幼叶cDNA文库的构建和鉴定 被引量:1

Construction and Identification of Young Leaf cDNA Library from Momordica charantia L.Infected with Fusarium solani

摘要 以白粉病菌侵染的苦瓜幼叶为材料,以λTrip IEx2为载体,利用SMART技术构建了1个由镰刀菌病原诱导后的苦瓜幼叶cDNA文库.结果显示,初始文库的滴度为4.0×10^6pfu·mL^-1,扩增后滴度为8.1×10^9pfu·mL^-1,插入片段大小分布在500~3000bp,平均值大于1000bp,重组效率为98%. To clone the powdery mildew resistance related gene from bitter melon (Momordica charantia L. ) , a cDNA library with cDNAs of bitter melon leaves infected with Fusarium solani was constructed by SMART (switching mechanism at 5' end of RNA transcript) and LD-PCR (long distance PCR) techniques. Results showed that the titers of unamplified and amplified cDNA library were 4.0 × 10^6 pfu . mL^-1 and 8.1 × 10^9 pfu . mL^-1 respectively, and the length of inserted fragments was 500-3000 bp,with an average length of more than 1 000 kb,and the recombination rate of 98%.
