摘要
目的探讨增殖细胞核抗原(PCNA)反义寡核苷酸对小鼠LA795肺腺癌皮下移植瘤生长的抑制作用。方法将20只接种LA795肺腺癌细胞T739小鼠随机分成两组。①治疗组接种LA795肺腺癌细胞后24h内用PCNA反义寡核苷酸(PCNA-ASODN)原位穿刺注射进行治疗;②对照组只注射Hank's液,每周1次,连续4周。观察两组大鼠肿瘤的一般情况与生存期。4周后断颈处死小鼠,完整地取下肿瘤,测量肿瘤体积,计算其抑瘤率。结果PCNA-ASODN治疗组小鼠LA795肺腺癌皮下移植瘤生长明显受到抑制,抑瘤率为68.53%。结论PCNA-ASODN对LA795肺腺癌皮下移植瘤生长具有抑制作用,通过反义技术可抑制靶基因的表达,从而抑制LA795肺腺癌细胞的增殖生长。
Objective To study the inhibitory effect of proliferating cell nuclear antigen antisense oligonucleotides ( PCNA - ASODN ) on the growth of lung adenocarcinoma LA795 in mice. Methods Twenty T739 mice bearing LA795 tumor cells were randomized into two groups. ①The treated group was given PCNA - ASODN once a week at 24 hours after cells inoculation. ②The control group was just treated with Hanks solution once a week for four weeks. The volume of tumor was measured. The inhibition rate of tumor growth was calculated. Results The tumor growth of PCNA - ASODN group ( 1 956 ± 138 mm) was significantly inhibited. The inhibition rate of tumor growth was 68.53%. Condusion It was demonstrated that the growth of LA795 tumor can be inhibited by PCNA antisense oligonucleotides, the expression of PCNA target gene and proliferation of LA795 tumor cells can be inhibited by PCNA antisense oligonueleotides.
