摘要
利用有机溶剂萃取法提取树舌灵芝MP-01发酵液中灵芝酸,并用分光光度法测定提取液中灵芝酸的含量,确定了分光光度法测定的最佳比色条件为:比色波长为520 nm,5%香草醛-冰醋酸和高氯酸用量分别为0.2 mL和1.0 mL(在反应系统中浓度分别为1.0%和83%),显色温度70℃,显色时间为25 min,在此条件下测得醇溶物和醇沉物提取液中灵芝酸含量分别为0.022 mg/g和0.049 mg/g。
Ganoderic acid produced during the growth of Ganoderma MP-01 in submerged culture was extracted using ethanol and chloroform, and its concentration was determined spectrophotometrically at 520 nm using oleonolic acid as the standard. Optimal assay conditions were: vanillin concentration 1%, perchloric acid concentration 83%, reaction temperature 70℃; reaction time 25 min. Ganoderic acid concentrations in SIE (substance insoluble in ethanol) and SSE (substance soluble in ethanol) fractions were 0. 049 mg/g and 0. 022 mg/g, respectively.
