摘要 目的:探讨内皮素-1对体外培养的正常人黑素细胞形态、增殖、酪氨酸酶活性及黑素合成的影响。方法:采用不含佛波醇酯和霍乱毒素的培养液培养黑素细胞,倒置显微镜观察细胞形态,分别用CCK-8法、多巴氧化法和NaOH法测定细胞增殖、酪氨酸酶活性和黑素含量。结果:内皮素-1可促进黑素细胞的树突形成,可上调酪氨酸酶活性,增加黑素合成,均在10nM时最显著。1~100nM的内皮素-1可明显刺激细胞增殖,随时间延长效应更明显。结论:一定浓度的内皮素-1具有促进培养的人黑素细胞树突形成、增殖、上调酪氨酸酶活性和增加黑素生成的作用。 Objective: To investigate the effects of endothelin- 1 on morphology, proliferation, tyrosinase activity and melanogenesis of normal human melanocytes in vitro. Methods: Melanocytes were cultured without phorbol esters and cholera toxin. Morphology was observed by invert microscope. The cell viability, tyrosinase activity, and melanin content were detected by CCK- 8, DOPA oxidase and NaOH methods, respectively. Results: Endothelin- 1 induced melanocyte dendricity and stimulated cell viability at the concentration of 1100 nM. It also increased tyrosinase activity and melanin content, with more marked effects at the concentration of 10 nM. Conclusion: Endothelin-1 exhibits stimulative effects on melanocyte dendricity, cell proliferation, tyrosinase activity, and melanin content at a certain coneentration.
