三个山羊群体抑制素-α 5′区的单核苷酸多态性分析 被引量:1

摘要 采用PCR-SSCP技术检测抑制素α(Inhibin-α,INHA)基因5′调控区在高繁殖力山羊品种(黄淮山羊和长江三角洲白山羊)以及低繁殖力品种(波尔山羊)中的单核苷酸多态性,同时研究该基因对黄淮山羊繁殖力的影响。设计3对引物扩增山羊INHA基因5′调控区序列,只有引物2在3个山羊群体中检测到多态性,出现了3种基因型(AA、AB、BB),测序结果表明BB型和AA型在INHA基因5′调控区都发生了2处碱基突变(260G→T和353 A缺失)。黄淮山羊突变纯合型(BB)和突变杂合型(AB)平均产羔数分别比野生型(AA)多1.25只(P<0.01)和0.78只(P<0.05)。初步表明,INHA基因可能是影响黄淮山羊高繁殖力的一个主效基因或是与其存在紧密遗传连锁的一个分子标记。 Single nucleotide polymorphisms (SNPs)of 5'regulatory region of the inhibin-α (INHA) gene were detected in high fecundity breeds ( Huanghuai goat and Yangtse River Delta white goat) and low fecundity breed( Boer goat) by PCR-SSCP. The effects of inhibin-α on Huanghuai goat fecundity were studied. The polymorphic loci were found by two pairs of pr/mers. Sequencing results revealed two mutations in 5'reg/on of INHA gene in all three breeds (260 bp →T and 353 bp A missing). The goats carrying homozygous mutation BB or heterozygous mutation AB had 1.36 ( P 〈 0.01 ) or 0. 86 ( P 〈 0. 05 ) in the number of lambs more than those with wild type AA in Huanghuai goat, respectively. These results pre- liminarily show that the INHA gene is a major candidate gene influenceing the prolificacy of Huanghuai goat or a potential molecular genetic marker closely linked to target gene (s).
