摘要
考察了5L发酵罐中三种甲醇流加策略(恒溶氧、恒比生长速率和恒甲醇浓度)对重组毕赤酵母表达人肝再生增强因子(hALR)的影响。控制恒定甲醇浓度为10g/L诱导时,甲醇比消耗速率和hALR表达速率显著高于其它两种流加策略,最终hALR表达水平为360mg/L。
The effects of three methanol feeding strategies (DO-stat, constant specific growth rate and methanol-stat) on the expression of human augmenter of liver regeneration (hALR) in recombinant Pichia pastoris were studied in a 5L bioreactor. When methanol concentration was controlled at 10g/L for induction, the specific methanol consumption rate and hALR production rate were significantly higher than those obtained with other strategies, and the final hALR expression level reached to 360mg/L.
