摘要
对库尔勒香梨中β-葡萄糖苷酶活性测定条件进行了研究。结果表明,粗酶液中添加2%PVP,在37℃条件下,加入pH=6.0柠檬酸-磷酸氢二钠缓冲液,反应时间为90min,于400nm波长条件下检测β-葡萄糖苷酶活性最为理想;在确定酶活条件的基础上,对气调贮藏、冷冻贮藏、涂膜保鲜贮藏条件下库尔勒香梨中β-葡萄糖苷酶活性变化规律进行了研究,结果表明,在不同贮藏条件下,库尔勒香梨中β-葡萄糖苷酶活性均呈下降趋势,贮藏条件在1%涂膜下酶活性最低,因此采用1%涂膜进行贮藏。
Determination of β-glucosidase activity in Kuerle pear was studied. The optimal determination conditions were as follows: adding 2% PVP and C4H2OT-Na2HPO4 buffer (pH = 6.0) into crude enzyme solution, then analyzing the enzyme activity with 400 nm wavelength at 37℃ after reaction for 90 min. Effects of the storage conditions on the enzyme activity were also investigated, results showed that, trader different storage conditions, including gas-processing, Frozen processing, Film processing, the enzyme activity showed a downward trend. 2% Film preservation showed to be the best for Kuerle pear storage due to the lowest β-glucosidase activity.
