摘要
用硫氰酸胍一步法提取小肠、肾脏和骨组织总RNA,用PstI和SmaI双酶切携带钙结合蛋白(CaBP)9K基因的pUC12质粒,获取CaBP9KmRNA在各组雌性大鼠中的变化。结果:卵巢切除组,小肠粘膜CaBP9KmRNA明显低于对照组;而卵巢切除后注射17-β雌二醇治疗组(0.5μg/100g体重)未出现降低。17-β雌二醇可影响小肠粘膜CaBP9K的基因表达。本结果提示,雌激素可能通过钙结合蛋白参与钙在大鼠体内的代谢。
Objective: To study the influence of 17βestradial on calbindin 9K gene expression in the ovariectomized rat intestines.Methods:(1)Extraction of total RNA from rat intestines kidneys and bones.(2)Expression of CaBP 9K mRNA in rat intestines by Northern blot with α 32Plabled CaBP 9K cDNA.Results: Decrease in CaBP 9K mRNA occurred in the intestines of the ovariectomized rats,but no decrease was found in those rats of the 17βestradial treatment group.Conclusion: Estrogen can influence CaBP 9K gene expression in the rat intestines.It is a possible way how estrogen regulates calcium metabolism in the rats.
