摘要
应用3H-TdR掺入和四甲基偶氮唑盐(MTT)法观察了天然与合成甾体激素对培养的人子宫动脉平滑肌细胞(HUASMC)增殖的影响。结果:雌激素对HUASMC增殖无明显影响,孕激素可促进HUASMC的DNA合成和细胞增殖(P<0.05),其作用呈剂量依赖性。雌孕激素联用比单用孕激素效果明显。合成的孕激素左旋十八甲基炔诺酮(LNG)和3-酮-地索高诺酮(DSG)促进HUASMC细胞增殖作用弱于天然孕酮(P<0.05)。睾酮对HUASMC增殖有较弱的抑制作用(P<0.05)。米非司酮对HUASMC增殖无影响,对雌孕激素的促增殖作用也无阻断效应。提示螺旋动脉的重建与孕激素有关,使用避孕药后螺旋动脉平滑肌增殖不良可能是导致不规则子宫内膜出血的重要原因之一。
Objeotlve:The aim was to study and compare the effective variation of natural and synthetic steroid hormones on cell proliferation of human uterine artery smooth muscle to investigate the pathogenic mechnism of abnormal bleeding following exogenous hormonal contraceptives. Design: In votro study on the cultured human uterine artery smooth muscle cell(HUASMC). Settlng: Department of Obstetrics & Gynecology, The First Teaching Hospital, Beijing Medical University Material:The samples were obtained from 5 women undergoing abdominal hysterctomy for benign gynecologic disease. None of the patients received hormonal treatment before operation. Methods: 3H TdR incorporation and MTT method. The student′ t test was used for analysis of data. Results: Oestradiol had no effect on proliferation of HUASMC.Both natural and synthetic progesterone promoted DNA synthesis and proliferation of HUASMC( P <0 05),but the effect of synthetic progesterone, LNG and DSG was weaker obviously than the effect of natural progesterone( P <0 05). Their proliferative effect was unreversible by mifepristone, which has no effect on proliferation of HUASMC. Testosterone could inhibit, slightly the proliferation of HUASMC. Concluson: These results showed that steroid hormones were involved in reconstruction of spiral arteries and the effect of natural progesterone is different from exogenous hormones. Bad proliferation and contraction of spiral arteries could be one of the important cause of irregular bleeding after using contraceptives.
