内毒素诱导犬急性肺损伤模型的建立 被引量:2

Establishment of acute lung injury model induced by endotoxin in dogs

摘要 目的建立犬内毒素性急性肺损伤模型。方法选择健康杂种犬8条,静脉注入LPS650μg/kg,当PaO2/FiO2<300时,可认为ALI模型复制成功;同时监测呼吸力学,采用放免法测定血TNF-α、IL-6和IL-10含量,电镜下观察肺组织病理改变。结果所有动物在16h左右出现急性肺损伤,表现为肺动态顺应性(Cdyn)、总顺应性(Ctot)下降,呼吸功(WOBvent)、吸气峰压(PIP)、吸气阻力(Rawi)上升,PaO2和PaO2/FiO2下降。血中TNF-α、IL-6和IL-10的浓度有明显上升,电镜下肺间质及肺泡炎性水肿出血、中性粒细胞浸润、Ⅱ型肺泡上皮细胞损伤。结论内毒素输注后16h左右达到ALI标准,且该模型稳定,重复性好。 Objective To establish acute lung injury (ALI)model induced by endotoxin in clogs.Methods Eight male mongrel dogs were injected LPS 650μg/kg via vein.When PaO2/FIO2〈300,the acute lung injury model was successfully established.We monitored respiratory mechanics,the contents of TNF-α, IL-6, IL-10 in plasma with RIA and observed pathology changes in lung of acute lung injury via electron microscope.Results All of dogs developed into acute lung injury model within 16 hours.It performed lung dynamic compli- ance,total compliance,PO2 and PaO2/FIO2 decreasing and breathing work (WOBvent),peak inspiratory pressure (PIP),inspiratory resistance increasing.It was founded the contents of TNF-α,IL-6,IL-10 in plasma increasing obviously.Interstitial lung and alveolar inflammatory edema bleeding,polymorphonuclear neutrophils infiltration, type 2 alveolar epithelial cell injury were clearly observed via electron microscope.Conclusion After endotoxin was injected about 16 hours,the model reached the standards of ALI.The model was very stable and repeative.
