摘要
实验以乙醇浓度、提取温度、提取时间和超声波功率等为考察因素,进行正交试验,建立起了经优化后的超声波辅助乙醇提取苦荞麦中总黄酮工艺,并采用高效液相色谱法,对苦荞麦中提取的总黄酮含量进行了测定。实验表明在色谱柱为Shim-pack VP-ODS柱(250 nm×4.6 mm,5μm);流动相为乙腈∶水∶乙酸(20∶76.67∶3.33);流速为0.6 mL/min;检测波长254 nm;柱温为30℃;进样量20μL的色谱条件下,测得苦荞麦中总黄酮含量为0.617%.
In this study we get the optimum conditions which used ultrasonic -assisted extraction to extract the TFH through the orthogonal experiments. And then we used HPLC to determinate the TFH. The chroma- tographic condition was as follows : analytic column : Shim - pack VP - ODS column ( 250 nm ×4.6 mm, 5 μm) ;The mobile phase was acetonitrile: water: acetic acid (20: 76.67: 3.33) ;The flow rate was 0.6 mL/min;UV detection wavelength was 254 nm;Column temperature was 30 ℃. By this method, the content of TFH from Tartarian Buckwheat was 0. 617%.
