非成分药材载体对反左金成分的亲和性初步研究

摘要 目的初步研究非成分药材载体对原中药有效成分的亲合性。方法将反左金全方中的各单味药以适宜方法除去原成分,制成非成分药材载体作为亲合色谱固定相。以醇水为流动相,对反左金方、黄连、吴茱萸醇提取液进行分离。以30 mL为一单位连续收集水样,用HPLC指纹图谱进行检测,并用总量统计矩法评价非成分载体固定相对药材提取液成分的亲和性。结果反左金液经过黄连非成分载体收集的第31~60 mL药液与吴茱萸提取液的总量中心矩非常接近,而黄连提取液经过吴茱萸非成分载体的第0~30 mL,31~60 mL药液与黄连样品的总量中心矩非常接近,吴茱萸提取液经过黄连固定的第0~30 mL药液与吴茱萸提取液总量中心矩较接近。结论非成分药材载体对相同来源成分的亲和性较大,而对非同源性成分亲和性较弱,因此可用作亲合色谱固定相构建中药复方多成分体系的分离方法。 Objective To study the affinities of medicinal material non-component carrier and active components of Chinese drugs. Methods The original components in each medicinal material of the counter-zuojin prescription was eliminated, and the affined chromatographic stationary phase was made from non-component medicinal carrier respectively. The components in counter-zuojin prescriptions, Radix Coptis and Fructus evodiae were separated with the alcoholwater as mobile phase. The alcohol-water emulsion was continuously collected each 30mL as a sample units, the affinities of non-component carrier stationary phase and components extracted from original materials was determined with fingerprint by HPLC and estimated with total quantum statistic moment. Results The central one-rank moment of counter-zuojin extract through Radix Copis stationary phase column was approximated very much to Fructus evodiae in 31st- 60th mL of the extract, the central one-rank moment of Radix Copis extract in 0-30th mL and 31st-60th mL through Fructus evodiae stationary phase column was approximated very much to that of Radix Copis sample, and the central one-rank moment of Fructus Evodiae extract through Radix Copis stationary phase column in 0-30th mL was approximated to Fructus Evodiae extract. Conclusion The affinities of non-component medicinal material carrier is more to the medicinal material in same source, but weaker to non-consanguinity components, which can be used as a stationary phase for separating many components system of traditional Chinese medicine.
