摘要
为了探讨米非司酮抗早孕作用是否影响了细胞外基质蛋白水解,作者应用免疫组织化学及原位杂交技术检测正常早孕及米非司酮流产蜕膜中tPA、uPA、PAI-1及uPA-R抗原及相应mRNA。结果表明uPA-R及其mRNA在米非司酮流产蜕膜组织的两种主要组分,尤其是腺体细胞中明显增加。提示米非司酮刺激uPA-R生成增加可激活细胞外基质蛋白水解,因而与抗早孕作用可能有关。
Objective: To investigete the effects of mifepristone on the localization and expression of tPA,uPA,PAI 1,and uPA R in human early decidua. Methods: The specimen of human early decidua obtained from women requesting termination of pregnancy of 6 ̄8 weeks gestation by curettage or treatment with mifepristone.The immunohistochemical techniques and in situ hybridization methods were used respectively to detect the antigens and mRNAs of tPA,uPA,PAI 1,and uPA R in decidual specimens. Results: The results revealed that specific staining for proteins and mRNAs of tPA,uPA,PAI 1,uPA R were observed in the cytoplasm of both decidual glandular and stromal cells.uPA R antigen and mRNA were markedly increased mainly in glandular cells in speciman from a woman receiving mifepristone. Conclusions: (1)The cellular localization and expression of tPA,uPA,PAI 1 and uPA R in human decidua may play an important physiological roles in maintenance of pregnancy.(2)The contragestetional effect of mifepristone may be mediated by promoting proteolysis of extracellular matrix via increase in synthesis of uPA R in decidua.
