摘要
为了获得精确的测量结果,满足进一步定量分析、活性分析的需要,利用反相高效液相色谱对微生物来源的醛糖还原酶抑制剂F01-195A进行了测试,考察了流动相种类、流动相配比、进样量、柱温、流动相流速五个因素对样品测试的影响。结果表明:流动相及配比、流动相流速对保留时间影响较大。最佳测试条件为:乙腈-水体系作为流动相,流动相配比为乙腈∶水=50∶50,柱温为35℃,进样量为5μL,流速为0.8mL/min。
In order to acquire accurate results to meet the requirement of further quantitative and active analysis, F01-195A, an Aldose Reductase Inhibitor from Metabolites of Microorganisms is analyzed by a reverse-phase high performance liquid chromatography (HPLC). How the mobile phase and its ratio, injection volume, chromatographic column temperature and flow rate affect the test is studied in detail. The results show that the mobile phase and its composition, as well as flow rate have significantly effects on retention time. The optimization operation eondition is that acetonitrile-water is used as the mobile phase and its volume ratio is 50 to 50, chromatographic column temperature is 35℃, injection volume is 5μL and flow rate is 0. 8mL/min.
