姜黄素对胶质瘤C6细胞垂体瘤转化基因下调作用的体外研究 被引量:3

摘要 目的探讨姜黄素对体外培养胶质瘤C6细胞垂体瘤转化基因(PTTG)表达的影响。方法体外培养胶质瘤C6细胞,给予不同浓度的姜黄素(10μmol/L、20μmol/L、30μmol/L)作用24h后应用半定量逆转录聚合酶链式反应(RT-PCR)检测PTTG mRNA表达情况,Western blot方法检测PTTG蛋白水平的变化。结果姜黄素作用后,C6细胞PTTG mRNA表达与PTTG蛋白水平均下降,差异有统计学意义(P〈0.01),且随着浓度的增加,其下降越明显。结论姜黄素具有下调胶质瘤C6细胞PTTG表达的作用,并呈剂量依赖性。 Objective To investigate the effect of curcumin on the expression of pituitary tumor transforming gene (PTTG) in glioma C6 cell line. Methods Glioma C6 cells were allocated into the control group and 3 curcumin treatment groups with curcumin treatment at 10, 20 and 30 μmol/L for 24 h. After the treatments, PTTG mRNA and protein expressions in the ceils were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting, respectively. Results The expression levels of PTTG mRNA in the 4 groups, showed significant differences between any two groups (P〈0.01). Significant differences were also found in PTTG protein expressions between the 4 groups of C6 cells after the treatment (P〈0.01). Conclusions Curcumin can down-regulate PTTG expression at both the mRNA and protein levels in glioma C6 cells in a dose-dependent manner.
