摘要
探讨了快速溶剂萃取法(ASE)提取黄连中生物碱的可行性,并比较了该方法与回流提取法和超声提取法的优越性。以黄连中盐酸小檗碱的提取率为指标,以高效液相色谱法(HPLC)为检测方法,用正交实验对快速溶剂萃取法从黄连中提取盐酸小檗碱的工作条件进行优化。最佳仪器参数:提取溶剂为80%乙醇+0.5%HCl,提取温度为130℃,静态提取时间为10 min,提取次数为1次。快速溶剂萃取法可作为黄连中生物碱分析测定的前处理方法。
In this study, the alkaloids were extracted from Coptis chinensis franch via accelerated solvent extraction (ASE), and the advantages of the developed ASE method were compared with traditional extraction methods such as ultrasonic extraction and reflux extraction. The extraction efficiency of berbefine hydrochlofide in Coptis chinensis franch samples determined by HPLC was used as the evaluation indexes, and the operational parameters of ASE were optimized by performing orthogonal experiments. The optimum conditions of ASE for extraction berberine hydrochloride in Coptis chinensis franch were as following: 80% ethanol (with 0.5% hydrochloric acid) as solvent, extraction temperature at 130℃, static extraction time of 10 min for one cycle. The ASE technique can be used to extract berberine hydrochloride in Coptis chinensis franch quickly and effectively, and it can act as the pretreatment method of the determination of alkaloid compounds in Coptis chinensis franch.
