人工湖底泥与水体真菌培养基的筛选 被引量:4

摘要 为定量研究淡水水域底泥和水体中的真菌种群分布,以广州暨南大学校内的南湖底泥及水体为研究对象,筛选出适合定量分析真菌的最佳培养基,采用平板计数法测定真菌数量.采用了酵母浸出粉胨葡萄糖琼脂培养基(YPD)、马铃薯葡萄糖琼脂(PDA)、察氏培养基(CA)、马铃薯琼脂培养基(PA)和混合培养基(HH)等5种培养基,分别按原配方、原配方中加入10%底泥浸出液、以及在原配方中不含抗生素的培养基中加入适量氯霉素等3种方法配制培养基.培养结果表明:各种培养基上生长的真菌数量和种类各异,同一种真菌在不同培养基上的生长情况也相差极大.加入底泥浸出液的5种培养基,无论是底泥还是水样,所培养出来的菌落数量多数都多于原配方;加入抗生素的两种培养基所培养出来的菌落数量都远远少于原培养基.研究结果说明加入10%底泥浸出液的酵母浸出粉胨葡萄糖琼脂培养基(YPD)更加适合用于底泥和水体真菌数量的调查. In order to investigate the population distribution of fungi in both the freshwater and sediments quantitatively, the South Lake in Jinan University was chosen as subject, and plate notation was used for measurement. Five culture medium of YPD, PDA, CA, PA and HH were employed for screening. For each medium, besides its original composing, two modified ones were also adopted. One modification way was adding 10% of sediment extracts and the other was adding proper quantity of Chloromycetin. The results showed that not only the species and quantities of fungi in these medium were different, but the same fungus on various cultures grew obviously differently. For either the sediments or the water sample, fungi densities on the medium with 10% of substrate sludge extract were greater than those on the original medium. While the fungi quantities growing on medium with Chloromycetin were less than those growing on original medium. It suggested that the medium of YPD adding 10% of sediment extracts were more suitable for fungal quantitative investigation in both the sediments and freshwater ecosystem.
