MN/CAⅨ基因在宫颈上皮内瘤变及宫颈鳞癌中的表达 被引量:3

Expression of MN/CAⅨ Gene in Cervical Intraepithelial Neoplasia and Cervical Squamous Cell Cancer

摘要 目的:探讨MN/CAⅨ基因在不同等级宫颈病变中的表达情况,其在宫颈病变的病情检测中的价值。方法:以我院妇科2005年6月至2005年10月阴道镜指引下活检确诊的患者86例为研究对象,其中慢性宫颈炎25例,CIN1级21例,CIN2级15例,CIN3级12例,宫颈鳞癌13例:取宫颈脱落细胞提取总RNA,行逆转录—多聚酶链反应(RT-PCR)观察MN/CAⅨ基因表达情况。结果:不同宫颈病变脱落细胞的MN/CAⅨ基因表达情况:宫颈炎12%,CIN1级76.19%,CIN2级86.67%,CIN3级91.67%,宫颈鳞癌92.31%,扩增出MN/CAⅨcDNA目的片段。CIN及宫颈磷癌患者的MN/CAⅨ基因表达率明显高于宫颈炎性病变者(P<0.001)。结论:MN/CAⅨ基因在CIN与宫颈癌中高表达,可用以鉴别宫颈瘤性病变及宫颈癌。宫颈脱落细胞MN/CAⅨ基因检测可以作为CIN病情监测的辅助指标。MN/CAⅨ基因可能成为一种有潜力的预测宫颈瘤性病变的肿瘤标记物。 Objective: To study the expression of MN/CAⅨ gene in the mutiple stages of cervical cancer, and evaluate the role of detecting the gene for the genesis of cervical cancer. Methods: The cervical scraping cells and biopsy tissue were collected for 86 patients, which consisting 25 cases of cervicitis , 21 cases of CIN1, 15 cases of CIN2,12 cases of CIN3, and 13 cases of squamous carcinoma of cervix. The presence of MN/CAⅨ was detected in all the cervical scraping cells using two-step RT-PCR. Resuits: The rates of the corresponding bands detected by RT-PCR in cervical scraping cells were 12% ,76.19%, 86.67%, 91. 67%, 92.31% in cervicitis , CIN1, CIN2, CIN3, and cervical cancer respectively . X^2 test indicated that there was significant difference between benign cervicitis and CIN / cervical cancer in the expression rate of MN ( P 〈 0.001 ). The positive expression rate of MN increased gradually with the grades of CIN increasing, but there were no significant difference among them( P 〉 0.05). Conclusion: MN/CAⅨ was highly expressed in CIN and cervical cancer. It is a useful marker for the differentiation between cervical neoplasia , cervical cancer and benign cervicitis MN/CAⅨ expression may correlate with progress of cervical neoplasia. The expression of the MN/CAⅨ detected in cervical scraping cells may be used as a potential biomarker for momitoring cervical neoplasia.
