摘要 以尖孢镰刀菌(Fusarium oxysporium)菌体可溶性蛋白及菌体分泌物抗原作为免疫原,制备了高效价的兔抗尖孢镰刀菌菌体可溶性蛋白及菌体分泌物抗原抗血清,提取并纯化了抗体IgG。通过优化ELISA反应条件,建立了检测黄瓜枯萎病的间接ELISA方法,检测的灵敏度很高;特异性试验结果表明,除与茄病镰刀菌有一定交叉反应外,与黄瓜上的其他病原菌均无交叉反应,由于尖孢镰刀菌引起的枯萎病和茄病镰刀菌引起的根腐病所施的农药相同,说明本方法具有较强的实用性;田间实际检测效果明显,早期表现症状的黄瓜检测结果均呈阳性,具有较高的准确度。 Soluble protein of Fusarium oxysporium and exoantigen was injected into rabbit as antigen, tiserum with high titer was acquired, and IgG was isolated and purified. Indirect ELISA was established to detect cucumber Fusarium wilt by optimizing ELISA reactive condition. This method could test with very high sensitivity. The test results for specialities showed that besides Fusarium solani( Mart. ) App. et Wr. , the IgG didn't have any cross reaction with other pathogens in cucumber. Owing to the fact that Fusarium wilt caused by Fusarium oxysporium and root rot by Fusarium solani ( Mart. ) App. et Wr. use the same pesticide, this method has good practicability and with distinctive effect in the fields. Cucumbers showed early symptom were all detected positive. This means the detection with very high accuracy.
