戊酸雌二醇对去势大鼠子宫雌激素受体亚型表达的影响 被引量:5

摘要 目的:观察戊酸雌二醇对去势大鼠子宫雌激素受体α(ERα)、ERβ表达的影响。方法:4月龄SD雌性大鼠80只,随机分为正常组、假手术组、去势组、药物组,每组20只。药物组大鼠于术后4周每天戊酸雌二醇灌胃给药,其余三组给予等量生理盐水灌胃,8周后处死大鼠,分离摘取子宫进行ERα、ERβ免疫组织化学染色及mRNA半定量RT-PCR检测。结果:(1)ERα、ERβ在正常子宫内膜腔上皮、腺上皮细胞表达较强,且ERα明显较ERβ表达强(t=2.713,P=0.014);(2)去势组ERα的表达较正常组、假手术组明显减弱(t=2.288,P=0.034);(3)药物组ERα的表达较去势组明显增强(t=5.308,P<0.01),ERβ表达无明显增强(t=1.505,P=0.150),ERα/ERβ比值升高(t=11.781,P<0.01)。结论:戊酸雌二醇可明显上调去势大鼠ERα的表达,但对ERβ的表达无影响。 Objective To investigate the effect of estradiol valerate on the expressions of estrogen receptor (ER) subtypes alpha and beta in the uterus of the ovarieetomized rats. Methods A total of 80 female SD rats were randomized to 4 groups: control group, sham-operated group, ovariectomied group, and drug group, 20 for each group. The drug group received daily intragastric estradiol valerate 4 weeks after ovariectomy, the other three groups received the same volume of normal saline. All rats were executed to obtain their uterus 8 weeks following administration. The expressions of ER-α and ER-β were detected in the uterus by immunohistoehemistry, so were the levels of mRNA expression of the two ER subtypes by semi-quantitative RT-PCR. Results Both ER-α and ER-β were highly expressed in the normal endometrial epithelium and the glandular epithelial cells, and the ER-α expression was more significant (P 〈 0.05). As compared with the control group and sham-operated group, ER-α expression was markedly declined in the ovariectomied group (P 〈 0.05). The expression of ER-α was significantly upregulated in the drug group than in the ovariectomied group (P 〈 0.05), whereas that of ER-β was not (P 〉 0.05). The ER-α/ER-β ratio was elevated (P 〈 0.01 ). Conclusion Estradiol valerate markedly upregulates the expression of ER-α but has no effect on that of ER-β in the ovariectomized rats.
