狂犬病病毒(Flury株)鸡胚低代毒(LEP)毒种的保存期试验研究及免疫原性测定

摘要 采用小鼠脑内接种法对保存不同年代的5批狂犬病病毒(Flury株)鸡胚低代毒(LEP)毒种进行病毒含量测定,并将每批毒种分别肌肉注射3月龄健康易感犬,21 d后采用小鼠脑内中和试验和荧光抑制试验分别测定犬血清抗体效价,评价毒种的免疫原性。结果显示,5批毒种每0.03 mL的病毒含量分别为104.67LD50、104.67LD50、103.5LD50、103.5LD50、104.17LD50;5批毒种免疫的10只犬血清抗体效价采用两种方法测定均达到1∶32以上,结果表明狂犬病病毒(Flury株)毒种在-70℃保存12年病毒含量仍符合标准规定,并保持有良好的免疫原性。 The virus content and immunogenicity of five batches master seeds of rabeis virus ( Flury strain, LEP) been stored for 2 - 22 years were determined by intracerebral inoculation to 11 -13 g Balb/C mice and intramuscular injection to 3 months Beagle dogs. The titers of all dogs' serum were determined by the methods of NIH and fluorescence inhibiting test. The results showed the virus content per 0.03 mL of five batches master seeds stored for different times at - 70 ℃ were 10^4. 67 LD50, 10^4.17 LD50 and 10^3. 5 LD50 respectively. The antibody titer of all dogs against rabies virus by intramuscular injection with five batches master seeds were more than 1: 32. Conclusions can be drawn that virus content of the master seeds of rabeis virus(Flury strain, LEP) stored at -70 ℃ for 12 years still measure up the regulations, and keep good immunogenicity.
