应用半重叠TCR γ特异性引物的PCR方法检测淋巴细胞白血病

摘要 采用更加敏感的半重叠式TCRγ特异性引物的PCR方法,对28例ALL初治、CR及BMT患儿进行检测,并用消化煮沸及酚抽提法分别对所有骨髓标本进行DNA提取,将PCR结果进行比较。结果表明:消化煮沸法用于微量标本的DNA提取优于酚抽提法,28例ALL患儿中16例检出TCRγ特异性条带,MRD组18例,阳性检出12例,其中免疫分型标记为B细胞型的4例,占25%,免疫标记为T细胞型者全部出现TCRγ阳性条带。并对该引物的语系特异性及双克隆重排问题进行了理论探讨。 PCR method of more sensitive semi-nested TCR γspecific primers was applied to detect 28 cases with acute lymphoblastic leukemia (ALL) in childhood. DNA of every bone marrow semples was extracted with the methods of digesting-biling and phenol-extraction, and their PCR results were compared. The results showed that the digesting-boiling method was superiour in DNA extraction of microspectimens to phenol-extraction method. 16 in 28 cases with ALL in childhood were PCR-TCR γgene rearrangement positive. 12 in 18 cases of MRD group were positive ,and 4 of them were Blineage by immunotype label (25. 0% ). All that immunotype label was T-lineage arised TCR γpositive. In this study we carried on discussion for lineage-specific of cell and both clonal gene rearrangement
