紫草多糖的免疫调节和肿瘤抑制活性研究 被引量:17

摘要 为研究紫草多糖在体外对小鼠脾淋巴细胞的免疫调节活性及对肿瘤细胞的细胞毒作用,本实验采用水提醇沉法,经脱蛋白、脱色、透析等方法提取紫草多糖,MTT法检测紫草多糖对淋巴细胞增殖及对HepG2、SPC-A-1肿瘤细胞的杀伤作用.实验结果显示:当质量浓度在121~364μg.mL-1时,紫草多糖能显著的促进ConA刺激的T淋巴细胞增殖(P<0.01),对HepG2肿瘤细胞有显著杀伤作用(P<0.01),对SPC-A-1肿瘤细胞有明显的杀伤作用(P<0.05),且随着浓度的提高,增殖作用或抑制作用增强.在质量浓度为364μg.mL-1时,对HepG2细胞的抑制作用明显高于对SPC-A-1细胞的抑制作用,而在质量浓度为24μg.mL-1时,对HepG2细胞的抑制作用低于SPC-A-1细胞,紫草多糖对HepG2细胞的抑制表现出更强的剂量-效应依赖性.以上结果表明,紫草多糖不仅有免疫调节活性,还对肿瘤细胞具有一定的细胞毒作用. The object of this study is to evaluate the immunoregulation and anti-tumor activities of Arnebia root poly saccbaride (ARPS) in vitro. The ARPS was obtained by hot-water extraction, alcohol precipitation, de-protein, decolor, and dialysis etc. from Arnebia root and was administered on mouse spleen lymphocyte and tumor cell with 24 〉g · mL^-1, 121μg · mL ^-1 , 364 μg · mL^-1, respectively. The proliferation rate of mouse spleen lymphocyte and the inhibition rate of tumor cell were studied to determine the effects of ARPS by methyl thiazolyl tetrazoliuml (MTT). The resuhs showed that ARPS could significantly promote the proliferation of T lymphocytes stimulating by ConA (P〈0, 01) when the ARPS concentration was controlled between 121 μg · mL^-1 to 364 μg · mL^-1 , and show significant cytotoxicity to HepG2tumor cell (P〈0.01) and SPC-A-1 tumor cell (P〈0.05). With the increase of concentration, the proliferation or inhibition increases. When the concentration of ARPS reached 364 μg · mL^-1 , the inhibition of HepG2 tumor cell was significantly higher than that of SPC-A-1 tumor cell , while at the concentration of 24 μg ·mL^-1 , lower than that of SPC-A-1 tumor cell. The inhibition effect of ARPS on HepG2tumor cell showed strong dose dependent effect. In summary, ARPS not only exerts the immunoregulation activities, but also has cvtotoxicitv to tumor cells in a certain extent.
