五倍子瘢痕膏水溶液对瘢痕疙瘩成纤维细胞增殖的影响

EFFECTS OF WUBEIZI OINTMENT SOLUTION ON PROLIFERATION OF KELOID-DERIVED FIBROBLASTS

目的探讨五倍子瘢痕膏治疗瘢痕疙瘩的作用机制。方法采取瘢痕疙瘩成纤维细胞原代培养,用四甲基偶氮唑盐比色法测定不同浓度药物对原代培养的第4-8代瘢痕疙瘩成纤维细胞增殖的影响。结果各不同浓度的药物组对瘢痕疙瘩成纤维细胞的增殖都有一定的抑制作用（P〈0.01）,并且有时间和剂量依赖。对体外培养的瘢痕疙瘩成纤维细胞周期研究表明,各药物组S期的细胞百分比显著高于对照组,其间有显著性差异（P〈0.01）;而各药物组G2＋M期细胞百分比却明显低于对照组,其间有显著性差异（P〈0.01）,并且这种对S期的阻滞作用和对G2＋M期细胞的抑制作用随药物浓度的增加而增强（P〈0.05）。结论不同浓度的五倍子瘢痕膏水溶液对瘢痕疙瘩成纤维细胞的增殖都有一定的抑制作用,其重要的作用途径是使瘢痕疙瘩成纤维细胞S期的细胞数上升,出现S期阻滞,分裂期的细胞数下降,从而抑制了瘢痕疙瘩成纤维细胞的增殖,并且这种抑制作用有一定的时间和剂量依赖。

Objective To evaluate the effect mechanism of Wubeizi Ointment （WBZ Ointment） on keloid. Methods Keloid-derived fibroblasts primary cells were cultured,the effects of the different concentration drugs on keloid-derived fibroblasts primary cell proliferation of passage 4-8 were examined by 3-（4,5-dimethyl-zthiahiazoyi）-3,5-di-plenyl-tetra zolium bromide （MTT） colorimetric method. Results Keloid-derived fibroblasts proliferation was restricted by each concentration drugs（P〈0.01） in a time and dose-dependent manner. The in vitro study of keloid-derived fibroblasts cell cycle suggested S stage cell proportion in each drag group was higher than that of control group. The difference was very significant（P〈0.01）;G2＋M stage cell proportion in each drug group was lower than that of control group. And the difference was very significant（P〈0.01）, the effect of blockade to S stage and restriction to G2 ＋M stage increased with concentration（P〈0.05）. Conclusion WBZ Ointment in different concentration have inhibitory effect on keloid-derived fibroblasts proliferation, by increasing the number of S stage cell and decreasing the number of mitotic stage cell.The restriction exerted in a time and dose-depandent manner.
