摘要
目的探讨大鼠额叶锐器损伤后细胞凋亡的变化规律及可能机制。方法采用大鼠额叶锐器损伤模型,用TUNEL法检测细胞凋亡,用RT-PCR和Western blot检测诱导性一氧化氮合酶(iNOS)基因和蛋白的表达变化。结果凋亡细胞在损伤后3h即可发现,24h达到高峰,随后逐渐减少;伤后3hiNOS基因和蛋白的表达开始上升,24h达到高峰,以后也逐渐下降。结论大鼠额叶锐器伤后存在细胞凋亡,凋亡细胞数量的变化与伤后时程有关,iNOS表达增加可能是影响细胞凋亡的重要因素。
Objective To investigate the alterations and possible mechanism of cell apoptosis after frontal sharp injury in rats. Methods Using frontal sharp cortex injury model, RT-PCR, western-blot and TUNEL methods were used to determine iNOS mRNA, iNOS protein and apoptotic cells respectively. Results The apoptotic cells were found as early as 3 hours after injury and reached a peak 72 hours, and then decreased in the following time. The iNOS mRNA and iNOS protein increased 3 hours after frontal injury, peaked at 24 hours, then reduced correspondently in the following time. Conclusion Cell apoptosis exists after frontal sharp injury in rats and the changes of the number of apoptotic cells relate with temporal course of post injury. The expression of iNOS increases may play a vital role in cell apoptosis.
