植物乳杆菌ST-Ⅲ产降胆固醇相关酶的研究 被引量:7

摘要 植物乳杆菌ST-Ⅲ菌株经验证具有降胆固醇的作用,在此基础上,对该菌株产生的和降胆固醇相关的酶进行了初步研究。实验发现,该酶为胞内酶,在培养基中加入胆固醇可诱导ST-Ⅲ合成降低胆固醇相关酶,添加量为2.5%时,酶活力最大;在粗酶液中加入Triton X-100可提高酶活力,在添加量为1%时达到最大值;硫酸铵提取该酶的最适饱和度为60%。实验还对该酶的酶活力测定方法进行了研究,确定了ATP以及底物的最适浓度分别为2.5mg/mL以及150μg/mL。 Lactobacillus plantarum ST-Ⅲ has been proved to be able to remove cholesterol. Based on the former experiments,explorative research has been done on the effectiveness of cholesterol removal by the enzymes which ST-Ⅲ produced. Experiment showed the enzyme was intracellular enzyme. The enzyme which produced by ST-Ⅲ could be induced by cholesterol in the fermentation medium, when the addition gets 2.5%, the enzyme activity was higher. Added Triton X-100 into the crude enzyme liquid could increase the enzyme activity, the optimal addition was 1%. 60% ammonium sulphate was the optimal precipitation of enzyme. And the activity determination of the enzyme was studied, the optimal concentration of ATP and substrates was 2.5mg/mL and 150μg/mL respectively.
