摘要
目的探讨胎鼠皮肤间充质干细胞的体外增殖特性。方法取昆明小鼠胎鼠皮肤体外培养胎鼠皮肤间充质干细胞,MTT法检测所培养细胞对表皮生长因子(EGF)、角朊细胞生长因子(KGF)和碱性或纤维细胞生长因子(bFGF)的反应,观察其在体外培养条件下的增殖与生长特征。结果胎鼠皮肤间充质干细胞具有很强的增殖活性,传代培养潜伏期为24-48 h,对数增殖期为3-5 d,然后进入平台期。经过15代连续培养的细胞其增殖能力未见明显改变。应用MTT法检测发现,培养的细胞对bFGF具有良好的反应性,并在1.0μg.L-1时反应最强,而对KGF和EGF的反应不明显。结论培养的胎鼠皮肤来源细胞增殖能力强;为间充质来源的干细胞。
Objective The characteristics of the proliferation of fetus mice skin MSCs were observed. Methods Fetus mice skin MSCs were purified through the method of cell culture in vitro and potential of proliferation were observed. Affections of bFGF, EGF and KGF on cells proliferation were observed by the method of MTT. Results The repeated adherent passage cultured cells that are originated from the fetus mice skin cells still had a strong proliferating capacity, after 15 generation' s passage purification culture. In our experiments, cultivating cells grew into lag phase 24 - 48 h after passage, and 3 - 5 d into logarithmic growth phase, then into platform phase. The adherent cultured cells have reaetivity to bFGF and the greatest effect dosage is 1.0μg/L, while cells no reaetibity to KGF and EGF. Conclusion It is confinned that the skin MSCs existing in the fetus mice have the potential of self-proliferation. It is clarified that the cultured cells are originated from stem cells derived dermis mesenchymal.
