应用PCR技术鉴定家鸡囊胚细胞性别的研究 被引量:2

摘要 为了鉴定家鸡囊胚细胞的性别,应用W染色体性别鉴定引物进行PCR,对不同方法提取的微量囊胚细胞DNA的效果进行鉴定,并应用此技术对种蛋进行性别鉴定。结果表明,3种方法均能提取适合于PCR的模板,其中以Chelex-100法提取的效果最好,最小细胞数为10个。对123个种蛋的性别进行鉴定,其中雌胚57个,雄胚66个。本研究对微量家鸡囊胚细胞DNA提取方法进行了大量的摸索和改进,为家鸡种蛋孵化前性别鉴定技术研究奠定了基础。 To identify chicken blastulan cell,W chromosome prime was used to identify the effect of mini-DNA extraction by PCR. The result showed that mini-DNA extracted by different method was for PCR template,and method of Chelex-100 was best for PCR with ten cell as template. We applied the technology to detect 123 eggs for sex identification. The results showed that there were 57 female blastulan and 66 male. The study developed the technology of mini-DNA extraction by different method,it was the base for sex identification in poultry egg without hatching.
