抗纤灵对5/6肾切除大鼠肾组织核因子κB及肿瘤坏死因子α,白细胞介素6 mRNA表达的影响

Effects of KangXianLing Recipe on the Expression of NF-κB and TNF-α mRNA,IL-6 mRNA of Renal Tissue in Rat Model of CRF

目的观察抗纤灵对5/6肾切除大鼠肾组织核因子κB(NF-κB)及肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)mRNA表达的影响。方法采用5/6肾切除法建立慢性肾衰大鼠模型,4周后按血肌酐水平将造模大鼠分为模型组和抗纤灵低、中、高剂量治疗组,8周后免疫组化法测定肾组织NF-κB表达;RT-PCR法测肾组织TNF-α,IL-6mRNA表达。结果模型组大鼠肾组织NF-κB及TNF-α,IL-6mRNA表达均较正常组明显升高(P<0.01);3个剂量治疗组与模型组比较均明显下降(P<0.01)。结论抗纤灵可通过降低模型大鼠肾组织NF-κB及TNF-α,IL-6mRNA表达延缓肾纤维化进展。

Objective To study the effects of KangXianLing recipe on the expression of NF - κB and TNF -αmRNA, IL - 6mRNA of renal tissue in rat model of CRF . Methods The rat model of CRF was established with 5/6 nephrectomy ,4 weeks later, the model group rats were randomly divided into pathologic group and KangXianLing low, middle and high dose treatment groups. After KangXianLing recipe was orally administered for 8 weeks , the expression of NF - κB and TNF -αmRNA, IL - 6mRNA of renal tissue were determined. Results Compared with normal group, the expression of NF - κB and TNF - αmRNA, IL -6mRNA in pathologic group were significantly increased （P 〈 0. 01 ） , and those in treatment gropes were obviously decreased （P 〈 0.01 ）. Conclusion KangXianLing Recipe may suppress fibrosis development, at least in part via down - regulating NF - κB and TNF - αmRNA, IL - 6mRNA expression.
