摘要
目的观察体外培养的骨髓内皮祖细胞(EPCs)在C反应蛋白作用后功能的改变,探讨C反应蛋白影响EPCs修复损伤血管内膜的可能机制。方法使用密度梯度离心法从骨髓获取单个核细胞并进行体外培养。对贴壁细胞进行细胞化学分析,以激光共聚焦显微镜鉴定FITC标记的荆豆凝血素I(FITC-UEA-I)和DiI一标记的乙酰化低密度脂蛋白(DiI-acLDL)双染色阳性细胞为正在分化的EPCs。加入不同浓度C反应蛋白,测定C反应蛋白作用后不同时间EPCs迁移、黏附、增殖能力。结果C反应蛋白作用后骨髓EPCs迁移、黏附和增殖能力明显减退。结论C反应蛋白可使骨髓EPCs迁移、黏附、增殖功能受损,且这种变化与C反应蛋白浓度与作用时间呈正相关。
Objective To investigate the effect of C-reactive protein (CRP) on the function of bone marrow-derived endothelial progenitor cells (EPCs) ,and study its possible mechanism. Methods Mononuclear cells were acquired from the bone marrow and cultured in vitro. Cytochemistry analysis was performed on attached cells after 7 days' culture. EPCs were characterized as DilacLDL/FITC-UEA-I double positive cell detected by laser confocal microscopy. EPCs were cultured with different concentrations of CRP (5,10,15 and 20μg/mL) for 12h,24h and 48h. The biological functions (abilities of immigration,adhension and proliferation) of EPCs were determined after culture. Results C-reactive protein significantly inhibited EPCs proliferation,migration and adhesive functions. Conclusion CRP may serve to inhibit the biological characteristics of EPCs. And these changes are positively correlated to dose and action time of CRP.
