摘要
运用大鼠佐剂性关节炎(AA)模型,牛磺鹅去氧胆酸(TCDCA)按0.2g/kg剂量,经灌胃给药后,测量致炎前和致炎后不同时期大鼠足跖肿胀度,并采用荧光分光光度法和ELISA双抗夹心法分别测定AA大鼠外周血中NO、LTB4含量。结果:TCDCA连续给药21d,可显著抑制AA大鼠不同时期的足跖肿胀,显著降低血清中NO水平;TCDCA连续给药4d,可显著降低AA大鼠外周血中LTB4含量。结论:TCDCA的抗炎作用与其抑制LTB4和NO产生有关。
To study on anti-inflammatory mechanism of TCDCA,Freund's complete adjuvant arthritis rats were used as a model of arthritis animals,and TCDCA was intragastric administrated. The paws swelling of AA rats were measured in different inflammatory periods. The contents of NO and LTB4 of peripheral blood were measured by fluorospectrophotometry and ELISA methods respectively in AA rats. The results indicated that after TCDCA were successively intragastric administrated for 21 d,TCDCA markedly inhibited AA rats hind paws swelling in different inflammatory periods,degraded the content of NO in blood serum. After TCDCA were successively intragastric administrated for 4 d,TCDCA degraded the content of LTB4 in peripheral blood remarkably. The anti-inflammatory actions of TCDCA have relationship with inhibiting the production of LTB4 and NO.
