摘要
目的研究丹七注射液中丹参素在犬体内的药物动力学。方法用高效液相色谱-紫外检测法,色谱柱:Lichrospher C18(4.6×250mm,5μm);流动相:乙腈-1%的冰醋酸水溶液(8∶92);流速:1.0mL/min;检测波长:280nm。结果丹七注射液犬静脉注射给药后,丹参素的经时血药浓度呈现快吸收慢消除的特点,丹参素在犬体内的动力学过程符合双室模型。其药物动力学参数为α=1.533h-1,β=0.5111h-1,T(peak)=0.25h,t1/2β=0.573h。结论丹参素在体内主要以消除过程为主,属慢消除过程,在体内作用时间持久。
Objective To study the pharmacokinetics of Danshensu (tanshinol) in Danqi Injection(DQI) in dogs. Methods HPLC - UV detection was used to determine the Danshensu level in biological samples. The Liehrospher C18 column(4.6×250mm,5μm) was used as an analytical column with a mobile phase consisted of acetonitrile - 1% acetic acid (8: 92), the flow rate being 1.0 mL· min^-1 and the wave - length being 280 nm. Results After intravenous injection of DQI in dogs, the plasma concentration - time curve of DQI in dogs fitted well to a two - compartment model. with the characteristics of fast absorption and slow elimination. Its pharmacokinetics parameters are as follows:α=1.533h^-1,β=0.5111h^-1,T(peak)=0.25h,t1/2β=0.573h. Conclusion The pharmaeokineties of DQI in dogs fits the two - compartment model. In the pharmacokineties of Danshensu, elimination course is the main course with the slow and lasting characteristics.
