“补肺肾祛痰瘀”膏方对支气管哮喘大鼠HPA轴和GR的影响 被引量:16

摘要 目的探讨"补肺肾祛痰瘀"膏方(简称"补膏")对哮喘大鼠HPA轴和GR的影响。方法建立SD大鼠哮喘模型,分为"补膏"高中低剂量组。从造模第15天起灌胃给药至末次激发后处死。分别用放射免疫分析法测定下丘脑CRH和血浆ACTH、CORT含量。SYBR GreenI染料法实时荧光定量PCR检测肺组织糖皮质激素受体(GR)mRNA含量。结果大鼠下丘脑CRH和血浆ACTH、CORT的含量变化基本一致,其中模型组含量比正常组明显降低(P<0.01);"补膏"低剂量组与模型组差异不明显(P>0.05);"补膏"中、高剂量组较模型组明显增高(P<0.01)。各组大鼠肺组织GRmRNA的表达,模型组最低,较正常组明显降低(P<0.01)";补膏"低、中、高剂量剂量组较模型组表达增多(P<0.05,P<0.01)。结论"补膏"主要通过温补肾阳,改善哮喘大鼠HPA轴功能低下状态,提高肺组织GRmRNA的表达,调节哮喘神经内分泌免疫网络紊乱,有利于哮喘的防治。 Objective To study the therapeutic effect of the "Bufeishen Qutanyu" herbal paste on the changes of hypothalamic-pituitary-adrenal (HPA) axis and glucocorticoid receptor(GR) in asthma rats. Methods The SD Rat model of asthma was made. The "Bufeishen Qutanyu" herbal paste groups included three groups: low-dose group, moderate-dose group and high-dose group. All rats were administered by warm water, low-, moderate- or high-dose "Bufeishen Qutanyu" herbal paste respectively from day 15 after the model was made to the last day of challenging by ovalbumin. Then the rats were sacrificed and their hypothalam tissue, blood samples and lung tissue were taken, the contents of CRH, ACTH and CORT were measured by radioimmunoassay and GR mRNA were measured by real-time PCR in all groups. Results In asthma model group, the concentrations of CRH, ACTH, CORT and GR mRNA of rats were decreased(P〈 0.01). Compared with model group, the concentrations of CRH, ACTH and CORT in low-dose group were not increased significantly (P〉 0.05): their concentrations in moderate- and high-dose groups were increased (P〈0.01); the concentrations of GR mRNA in low-, moderate- and high-dose groups were increased (P 〈 0.01). Conclusion By warming kidney yang, "Bufeishen Qutanyu" herbal paste can regulate the function of neuroimmunoendocrine network, up-regnlate the function of HPA and expressional levels of GR mRNA in lung tissue of asthma rats, eventually to treat asthma.
