摘要
目的:建立川产九节龙药材中九节龙皂苷Ⅰ的HPLC-ELSD测定方法,考察了药材中九节龙皂苷Ⅰ含量。方法:采用Phenomenex C_(18)色谱柱(4.6 mm×250 mm,5μnm);流动相为甲醇-水(75:25);漂移管温度为73.8℃,载气流量为2.0 L·min^(-1)。结果:九节龙皂苷Ⅰ能与其他成分良好分离,药材中九节龙皂苷Ⅰ含量差异较大,九节龙皂苷Ⅰ在0.88~4.42μg范围内,线性关系良好(r=0.9999);样品平均同收率为99.8%,RSD为1.3%(n=6);结论:本ELSD-HPLC方法可有效测定川产九节龙中九节龙皂苷Ⅰ的含量。
Objective : To establish an HPLC - ELSD method to determine the contents of ardipusillosideⅠ in Ardisia pusilla A. DC. in different growing periods ( in February, May and August). Method : An HPLC - ELSD method was developed. A Phenomenex C18 (4.6 mm× 250 mm,5 μm)column was used , and the mobile phase consisted of methanol - water(75: 25). Flow rate was 1.0 mL · min^-1 at room temperature. Temp tube :73.8 ℃, Gas flow : 2.0 L · min^-1. Results:Ardipusilloside Ⅰ could be well separated from other compounds of A. pusilla,its contents in A. pusilla was different in various growing periods. The contents of ardipusilloside Ⅰ in A. pusilla collected in February was higher than that in other periods. Ardipusilloside Ⅰ showed good linearity in the range of 0. 88 - 4.42 μg , the average recovery was 99.8% ( n = 6), with RSD of 1.3 % ( n = 6). Conclusion: The HPLC - ELSD method is suitable for ardipusilloside Ⅰ in A. pusilla.
