摘要 目的:以大孔树脂-凝胶柱层析法制备羟基红花黄色素A(HSYA)。方法:室温浸提法提取红花水提液,大孔树脂柱层析法制备HSYA粗品,Sephadex LH-20凝胶柱层析法制备HSYA纯品,HPLC法分析HSYA纯品纯度,观察该HSYA抑制ADP诱导家兔体外血小板聚集的作用。结果:红花水提液经大孔树脂-凝胶柱层析法获得HSYA的纯度为95.3%,该成分抑制ADP诱发的家兔血小板聚集呈量效关系。结论:大孔树脂-凝胶柱层析法制备HSYA的方法为一简便、有效的技术。 Objective: Preparing Hydroxysafflor Yellow A (HSYA) by macroporus resin-gel column chromatography. Method:Aqueous extract was prepared from Carthamus tinctorius L.. Crude HSYA was prepared by macroporus resin chromatography and purified HSYA was further prepared by Sephadex LH-20 column chromatography. The purity was analyzed by HPLC. The inhibition of HSYA on rabbit platelet aggregation induced by ADP was observed. Result: From aqueous extract HSYA was prepared through macroporus resin-gel column chromatographic procedure and its purity was 95.3 % . This ingredient was active to inhibit platelet aggregation induced by ADP. Conclusion: HSYA preparation by macroporus resin-gel chromatography is a simple and effective technology.
