罗格列酮对糖尿病大鼠心肌组织中磷酸化IKK,IκBα表达的影响

摘要 目的探讨罗格列酮对链尿佐菌素STZ糖尿病大鼠心肌组织中核因子(NF)-κB抑制蛋白激酶(IKK)和其下游作用物NF-κB抑制蛋白(IκBα)表达的影响。方法将48只雄性Wistar大鼠随机分为正常对照组(A)、糖尿病组(B)和罗格列酮干预组(C),分别于实验8周和12周检测血糖、血清C肽水平,心重指数和心肌细胞面积,并用免疫组化方法检测8周和12周STZ大鼠心肌组织中磷酸化IKK和磷酸化IκBα表达的改变。结果与正常对照组相比,糖尿病组和罗格列酮干预组的心重指数、心肌细胞表面积及IKK和IκBα表达增加(P<0.01);而罗格列酮干预组的心重指数、心肌细胞表面积及IKK和IκBα表达低于糖尿病组(P<0.01)。讨论罗格列酮能抑制IKK活化,进而抑制IκBα降解,降低NF-κB活化水平,最终延缓心肌损伤,起到心肌保护作用。 Objective To investigate the mechanism of heart protective effect of rosiglitazone on diabetic rat heart.Methods 48 male rats were randomly divided into normal control group (A),diabetic rat group (B) and rosiglitazone treatment group (C). After 8 weeks and 12 weeks, immunohistochemistry were used to examine the expression of IKK and IκBα phosphorylated in the heart of STZ diabetic rats. And blood glucose, weight, index of heart weight, area of heart cell and C peptide in the blood were measured at the same time. Results In comparison with group A, the expression of IKK and IκBα phosphorylated in the heart of group B and group C was markedly increased ;and the index of heart weight, area of heart cell were bigger than group A(P〈0.01).Compared with Group B,the expression of IKK and IκBα phosphorylated in Group C was significantly lower ;and the index of heart weight ,area of heart cell were smaller (P〈0.01). Conclusions Rosiglitazone may play a protective role in the Heart of STZ Diabetic Rats by down-regulating IKK and IκBα and ultimately inhibiting NF-κB activation.
