摘要
目的探讨甲状腺过氧化物酶(TPO)mRNA在甲状腺癌和良性甲状腺结节中的表达情况及意义。方法应用逆转录聚合酶链反应(RT-PCR)技术,对55例甲状腺手术新鲜标本的TPOmRNA丰度进行检查。用光密度扫描仪测定并计算样品的TPO/β-actin比值,代表TPOmRNA丰度的相对含量。结果甲状腺组织中良性病变TPOmRNA阳性率93.94%%,恶性结节TPOmRNA阳性率54.55%,两者的阳性率比较(P<0.05)有统计学意义,恶性结节TPOmRNA的表达率低于良性病变。表达TPOmRNA病例的半定量观察结果,良、恶性病变组间TPOmRNA的光密度积分值(mRNA丰度)差异具有统计学意义(P<0.01),良性病变组明显高于恶性病变组。结论TPOmRNA在甲状腺病变的表达可视为良性病变的标志,在良恶性甲状腺病变的鉴别中具有重要应用价值。
Objective to explore the expression of the thyroperoxidase(TPO) mRNA in thyroid diseases and implication. Methods Abundance of TPO mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR) technique in the samples from 55 thyroid masses. Results TPO mRNA was detected in 93.94% of benign thyroid diseases and in 54.55% of the thyroid carcinomas, demonstrating a significant difference in TPO expression between the two thyroid diseases (17〈0.05). Semiquantitative analyses of the patients expressing TPO mRNA revealed that there was statistical difference between benign and carcinoma groups (117〈0.01). Ratio of the TPO/β -actin in benign groups was higher than in malignant groups. Conclusion Conclusion the expression of the TPO mRNA in thyroid diseases can be considered that it is the marker of the benign thyroid diseases. TPO mRNA was important valuable in diagnosing thyroid diseases.
