摘要
[目的]评价TTNPB对人体PBMC和T淋巴细胞的增殖的影响,确定剂量水平,建立一套通过受体途径研究维生素A功能的实验方法。[方法]用密度梯度分离法和磁性细胞分选法从健康人的外周血分离PBMC和T淋巴细胞,体外培养,采用MTT法,绘制人PBMC和T淋巴细胞的体外生长曲线,并测定在10-2mol/L,10-3mol/L,10-4mol/L,10-5mol/L,10-6mol/L,10-7mol/L6个不同浓度时的OD值,用方差检验分析各剂量对细胞的影响。[结果]TTNPB在10-2mol/L,PBMC的OD值为0.0600±0.0328(P﹤0.05),T淋巴细胞的OD值为0.0907±0.0110(P﹤0.05)。[结论]TTNPB在高浓度10-2mol/L对PBMC和T淋巴细胞增殖有显著的抑制作用;中等浓度有促进作用,但这还需要进一步的实验得出;低浓度时没有抑制或促进作用。
[Objective] To evaluate the effects of TTNPB on proolifcration of PBMC and T-Lymphocytes of human, and to identify the dose status as well as establish a empirical method from RAR- and RXR-ligand to study the functions of vitamin A. [Methods] PBMC and T-Lymphocytes were separated from the blood of healthy person by the way of the density gradient separation and MACS, then cultured in vitro. MTT method was used to draw the growth curve and measure the OD value at different concentrations of 10^-2 mol/L, 10^-3 mol/L, 10^-4 mol/L, 10^-5 mol/L, 10^-6 tool/L, and 10^-7 mol/L, respectively. Then One- Way-ANOVA was used to analyses the effect of different concentrations on cells. [ Results] A concentration at 10^-2 mol/L of TTNPB made OD value 0.060 0 ± 0.032 8 of PBMC (P 〈 0.05), and 0.090 7± 0.011 0 of T-Lymphocytes. [Conclusion] A high concentration at 10^-2 mol/L of TTNPB significantly inhibited the proliferation Of PBMC and T-Lymphoeytes, and more experiments was needed to made to statement that the middling concentration enhance the proliferation, the low concentration has no effect.
