摘要
[目的]探讨白藜芦醇(resveratrol,Res)对淋巴瘤Raji细胞增殖及细胞因子白介素-8(IL-8)和血管内皮细胞生成因子(VEGF)基因表达和分泌的影响。[方法]用不同浓度白藜芦醇处理Raji细胞48h后,用MTT法检测细胞增殖指数,用ELISA法检测IL-8和VEGF蛋白含量,用逆转录/聚合酶链反应(RT/PCR)检测IL-8和VEGF mRNA表达情况。[结果]白藜芦醇体外对Raji细胞无增殖抑制作用,不同浓度白藜芦醇作用Raji细胞48h后,白藜芦醇1组、2组和3组IL-8、细胞上清液VEGF蛋白含量,均显著低于对照组(P<0.05);IL-8、VEGF mRNA表达相对强度,均显著低于对照组(P<0.05),且随浓度增加而减弱。[结论]白藜芦醇体外对Raji细胞无增殖抑制作用,但可抑制IL-8和VEGF的分泌及mRNA的表达,这可能是白藜芦醇抗肿瘤血管生成的重要机制。
[Objective] We aimed to investigate the effects of angiogenic inhibitor vitro. [Method] We examined the serum contents of IL-8 and VEGF in supernatant, by specific ELISAs of Raji cells after treatment by Res of different dosage(25umol/1-100umol/l). The expression of IL-8 and VEGF mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). [Results] The protein contents of IL-8 in supernatant of Res(25umol/1-100umol/l) were(532.3 4- 54.6)pg/ml, (487.3± 63.0) pg/ml and (380.9 ±47.9) pg/ml respectively, significantly lower than that in the control. The protein contents of VEGF in supernatant of Res (25umol/1-100umol/l) were (73.3 ± 10.9) pg/ml, (46.6 ± 19.8) pg/mt, (30.2 ± 12.8) pg/ml respectively, significantly lower than that in the control. The relative values of expression of IL-8 mRNA in treated groups (25umol/1-100umol/l) were (0. 21± 0.06 ), (0. 17±0.09) and (0. 12±0.07) respectively. The values of expression of IL-8 mRNA were significantly lower than control group. And the values were in inverse correlation with concentration. The relative values of expression of VEGF mRNA in treated groups were (0.46± 0.21), ( 0.21 ± 0.13) and ( 0.09± 0.08) respectively. The values of expression of VEGF mRNA were significantly lower than control group and were in negative correlation with concentration. [Conclusions] These findings suggest that Res may inhibt the expression and secretion of IL-8 and VEGF, which may be one of the most important mechanisms.
