摘要
目的建立小鼠MHC-Ⅱ类分子表达水平检测的RT-PCR方法。方法采用RT-PCR法检测昆明小鼠H-2IAb基因表达水平,同时运用流式细胞术法对小鼠淋巴细胞表面H-2IAb的表达进行定量检测;比较两种检测方法间的相关性。结果RT-PCR电泳结果显示所有小鼠均检测出H-2 IAb基因;流式细胞仪检测结果显示所有小鼠淋巴细胞表面均有H-2IAb的表达,同时与RT-PCR检测结果相关。结论RT-PCR法对小鼠MHC-II类分子的表达水平进行检测条件可行,方法成立。
Objective To establish RT-PCR method to detect Mouse MHC-Ⅱ molecules expression. Method The H-2 lab gene was detected with the technique of one step reverse transcription Polymerase Chain Response (RT-PCR), also, the expression of H-2 lab in lymphocyte was tested by Flow cytometry. Study the relationship between the two results. Result The RT-PCR eleetrophoregram showed that all of the mice express the H-2 lAb gene. The expression (Mean.) of H-2 lAb with flow cytometry could be tested in all mice, which correlated with the result of RT-PCR( r = 0.649). Conclusion The RT-PCR method can be used to measure the expression level of Mouse MHC-Ⅱ molecules.
