摘要
目的观察酪氨酸酶相关蛋白(TRP)诱导实验性自身免疫性葡萄膜炎(EAU)大鼠调节性T淋巴细胞及其相关细胞因子的变化,探讨其可能的免疫机制。方法Lewis大鼠24只,随机分为实验组(10只)、Freund佐剂组(7只)和空白对照组(7只)。TRP100μg腹腔内注射Lewis大鼠,建立动物模型。注射后第15d取动物外周血流式细胞学三色荧光测CD4+、CD25+、Foxp3阳性细胞比例,ELISA法测血清中IL-10、TGF-β质量浓度行统计学比较。结果炎症期实验组动物外周血中CD25+细胞比例升高,Foxp3阳性细胞比例明显下降(P<0.01);血清中IL-10、TGF-β质量浓度下降(P<0.01)。结论EAU炎症期外周血T淋巴细胞活化;调节性T淋巴细胞(Foxp3阳性细胞)和细胞因子(IL-10、TGF-β)减少致免疫抑制作用减弱,参与了葡萄膜炎的发生和发展。
Objective Uveitis is recognized as an organic-specific autoimmune disease associated with T lymphocyte infiltration.Our study was to observe the changes of regulatory T cells and relevant cytokine in Lewis rats with experimental autoimmune uveitis(EAU) induced by tyrosinase related protein(TRP) and explore the possible effect of T lymphocyte in immunological mechanism of EAU. Methods Twenty-four Lewis rats were divided into 3 groups.100 μg TRP+0.1 mL Freund’s adjuvant+0.1 mg/0.1 mL of pertussis toxin was injected intraperitoneally to induce EAU in 10 rats as experimental group.0.1 mL Freund’s adjuvant+0.1 mg/0.1 mL of pertussis toxin was injected intraperitoneally in 7 rats as Freund’s adjuvant group,and 0.2 mL physical salt solution was used in 7 rats as blank control group.CD4+,CD25+,Foxp3+ T lymphocytes in peripheral blood were detected by Flowcytometry,and IL-10,TGF-β in serum were detected by ELISA at the fifth day after injection. Results The mean level of CD25+ T lymphocytes in rat peripheral blood was 1.50%,0.41%,0.44% respectively,and Foxp3+ T lymphocytes in rat peripheral blood was 22.39%,54.96%,62.03%,respectively in the three groups.The mean level of IL-10 in rat serum was 38.80 pg/mL,81.66 pg/mL ,78.56 pg/mL and that of TGF-β was 17.47 ng/mL,22.68 ng/mL,24.04 ng/mL in the three groups.Compared with Freund’s adjuvant and blank control group,CD25+ T lymphocytes were significantly ascended but Foxp3+ T lymphocytes,IL-10 and TGF-β were significantly descended in experimental group(P〈0.01).There were no statistically significant differences in above indexes between Freund’s adjuvant and blank control group(P〉0.05). Conclusion T lymphocytes are activated during EAU.Destruction of immunological tolerance caused by decrease of regulatory T cells and relevant cytokine may be involved in the occurrence and development of EAU.
