中华眼镜蛇毒因子对小鼠心脏移植急性排斥反应及MHC-Ⅱ和B7影响

摘要 目的探讨中华眼镜蛇毒因子(CVF)消耗补体对小鼠同种心脏移植急性排斥反应及MHC-Ⅱ和B7表达的影响。方法建立Balb/c至C57小鼠颈部同种异体心脏移植的动物模型,CVF实验组经静脉注射中华眼镜蛇毒因子以消耗受体血清中补体,观察移植心存活时间,并从CVF实验组和对照组中分别抽取12只小鼠于术后3、5和7 d定时处死,对比观察移植心急性排斥反应程度,以及MHC-Ⅱ、B7-1和B7-2的表达情况。结果CVF实验组,其移植心存活时间显著延长,平均达(26.2±s 1.7)d,与对照组(8.4±0.4)d相比有非常显著的差异(P<0.01);移植心肌组织病理检查结果,CVF实验组病变明显较对照组轻(P<0.01);MHC-Ⅱ类分子和B7-1、B7-2共刺激分子总体表达及单个细胞表达值CVF实验组较对照组显著减少(P<0.01)。结论中华眼镜CVF清除受体血清中补体可抑制小鼠同种心脏移植急性排斥反应,降低MHC-Ⅱ、B7-1、B7-2的表达,显著延长移植心存活时间。 AIM To investigate the effects of complement depletion with cobra venom factor (CVF) on acute rejection and expression of MHC- Ⅱ , B7 after cardiac allotransplantation in the mouse. METHODS The Balb/c-C57 mouse models of heterotopic cardiac allograft were developed and divided into two groups, control group and CVF-treated group. The survival time of the allograft was observed. Twelve mice chosen from the two groups respectively were killed at d 3, 5, 7, and the pathology grade for acute rejection, the expression of MHC- Ⅱ B7-1, and B7-2 was compared. RESULTS The survival time of cardiac allograft was (26.2 ±s 1.7) d in CVF-treated group and (8.4 ± 0.4) d in control group with a significantly difference (P 〈 0.01 ) . The grafts of CVF-treated mice showed the pathology grade for acute rejection and the expression of MHC- Ⅱ , B7-1, B7-2 were significantly decreased (P 〈 0.01) . CONCLUSION Cleaning up the complement in the serum with CVF can inhibit the acute rejection in the mouse, significantly prolong the survival time of cardiac allograft, and decrease the expression of MHC-Ⅱ , B7-1, B7-2.
