摘要
目的:通过分析炎性细胞因子TNF-α mRNA的变化,探讨参芎滴丸与环磷酰胺对急性脑梗死炎性反应的影响。方法:用电凝法闭塞大鼠大脑中动脉制成大脑中动脉闭塞(middle cerebral artery occlusion,MCAO)模型,于造模后12、48、72h三个不同时点处死大鼠,取梗死区脑组织备用,采用RT-PCR方法观察不同剂量参芎滴丸与环磷酰胺对缺血大鼠脑组织TNF-α mRNA表达的影响。结果:模型组大鼠TNF-αmRNA表达明显升高,与正常组、假手术组比较差异有显著性(P<0.05);参芎滴丸低(5倍)、中(10倍)、高(20倍)三个剂量组、环磷酰胺组梗死后24、48、72h三个不同时点TNF-αmRNA的表达明显下降,与模型组比较差异有显著性(P<0.05);不同剂量组之间差异有显著性(P<0.05),呈现量效关系;高剂量参芎滴丸的作用强度与环磷酰胺相似。结论:参芎滴丸与环磷酰胺均能显著抑制MCAO模型大鼠TNF-α mRNA表达,进而抑制梗死早期的炎性反应,高剂量参芎滴丸的作用强度与环磷酰胺相似。
Objective:To investigate the effect of "Shen Xiong Diwan"(SXD)and cyclophosphamidum(Cytoxan)on acute cerebral infarction inflammation response by detecting the changes of tumor necrosis factor-α(TNF-α) in brain after cerebral ischemia.Methods:Rat mode of middle cerebral artery occlusion(MCAO)was made with middle cerebral artery occlusion by electrocoagulation method.SXD and Cytoxan were given 8 days before operation,one time per day.MCAO rat model was divided into 4 groups randomly as following:high dose SXD group,middle dose SXD group,low dose SXD group,and cytoxan group.Sham operation group,model group,and normal group were used as control groups.The samples were collected 24 hours,48 hours and 72 hours after operation respectively.The expression of TNF-α mRNA was detected by RT-PCR.Results:The expression of TNF-α mRNA increased significantly in model group compared with that in normal and sham operation groups,and it was dramatically lowered by SXD.The effect of the high dose SXD was similar to that of Cytoxan.Conclusion:The results indicate that SXD and Cytoxan can inhibit inflammation response of MCAO model rat in early stage of the infarct through inhibition of TNF-α mRNA expression.
